The primary objective of the work was to develop an indirect ELISA assay for the detection of Crimean-Congo Haemorrhagic fever (CCHF) virus. Recombinant nucleoprotein from Crimean-Congo Haemorrhagic Fever (CCHF) virus was successfully derived from a bacterial expression system and purified for use in a novel enzyme-linked immunosorbent assay (ELISA) diagnostic test. ELISA tests were performed for detection of IgG antibodies, thus allowing efficient detection of antibodies. The major benefits of the assay also included removing any requirement for polyclonal sera, thus eliminating variation in preparations and allowing standardization between laboratories. The assay was successfully tested using a panel of positive sera supplied from samples identified as being positive. It is envisaged that this simple diagnostic ELISA for CCHF virus infection which removes the reliance on polyclonal antibody preparations, will be accessible to a wider range of laboratories enabling them to carry out routine diagnosis. This will improve the efficiency of diagnosis and subsequent management of infected animals. The dried fruits (500 g) of Termanalia chebula were pulverized and grinded. The grinded powder was then used for isolating an active component of it, chebulinic acid. 12-week-old(3 month) male Sprague-Dawley (SD) rats (n = 28) with initial body weights of 200-220g were taken from National Animal Laboratory Center, Central drug research institute (CDRI) To assess the therapeutic effect of chebulinic acid on Benign Prostatic Hyperplasia (BPH), rat BPH model has been successfully generated by testosterone induction method. normal control group was set up,. After the pre-treatment of 28 days, the prostate was dissected out to carry histological studies in order to confirm the establishment of model. Then BPH induced rats were randomly divided into four groups, and the rats those did not receive TP treatment became the normal control group. Thus, the rats were divided into five groups with 4 animals in each group: (a) a normal control group that received ethanol with edible oil; (b) a BPH group that received TP soluble in edible oil (c) a positive control group that received finasteride (Fi) (1 mg/kg), a 5ARI which is frequently used as a treatment for BPH , with TP (5 mg/kg); and (d) a group that received Chebulinic acid (50 mg/kg) with TP (5 mg/kg).(e) a group that received Chebulinic acid (100 mg/kg) with TP (5 mg/kg), and then for assessing the effect of Chebulinic acid Prostate weight index calculation was done of all group of rats along with Histopathical examination. Treatment with Chebulinic Acid significantly suppressed the development of prostatic hyperplasia and the prostatic weight was significantly reduced by Chebulinic Acid and histological indices were affected. Histopathological examination of BPH rats revealed glandular hyperplasia of the prostate accompanied with numeral secretions, whereas the rats treated with Chebulinic Acid showed mild glandular hyperplasia. The result suggests that Chebulinic Acid might inhibit the development and progression of BPH induced by Testosterone. In conclusion, the present study showed that administration of Chebulinic Acid to BPH rats decreased the prostate weight and restored the altered histological architecture of the rats. Hence, from a clinical point of view, Chebulinic Acid could have applications in elderly subjects with BPH. Bachelor of Science (B.Sc) in Biotechnology, Chemistry and Zoology from, Govt.
